EXPERIMENTAL BASIS FOR PREVENTING GASTROINTESTINAL INFLAMMATION IN RATS EXPOSED TO POTASSIUM PERCHLORATE

Background. The accumulation of perchlorates in Ukraine during active combat operations makes it important to find ways to prevent the harmful effects of perchlorates. Based on the established antioxidant and anti-inflammatory properties of quertulin, propoxazepam and their combination, it was logical to confirm these effects of the complex on a model of prolonged potassium perchlorate intoxication. Therefore, the aim of the study was to investigate the anti-inflammatory, antimicrobial and antioxidant effects of propoxazepam and quertulin in the mucous membranes of the digestive tract of rats under conditions of prolonged potassium perchlorate administration.
Methods. The study was conducted on 7-month-old female rats, which were divided into 4 groups: 1 – intact; 2 – potassium perchlorate; 3 – potassium perchlorate + 10 μg/kg L-thyroxine; 4 – potassium perchlorate + 400 mg/kg quertulin + 200 μg/kg propoxepam. The potassium perchlorate solution was administered to the rats in drinking water, while L-thyroxine, quertulin and propoxepam were administered orally on a daily basis. The duration was 40 days. After removing the rats from the experiment, markers of inflammation (acid phosphatase and elastase activity), bacterial contamination (urease activity) and lipid peroxidation (malondialdehyde content) were determined in the homogenates of the mucous membranes of the oral cavity, stomach, small and large intestines.
Results. Long-term administration of potassium perchlorate to rats with drinking water caused an increase in inflammation markers (acid phosphatase, elastase), an outbreak of lipid peroxidation (malondialdehyde content) and increased microbial contamination (urease activity) in the mucous membranes of the digestive tract of animals. Prophylactic administration of L-thyroxine to rats under conditions of potassium perchlorate intoxication only partially prevented the established disorders in the mucous membranes of the digestive tract of rats, which indicates the direct damaging and inflammatory effect of potassium perchlorate, which is not related to the function of the thyroid gland. The combination of quertulin and propoxazepam effectively eliminated the manifestations of perchlorate intoxication: it prevented the increase in markers of inflammation, lipid peroxidation and microbial contamination in the mucous membranes of the digestive tract of rats, bringing them closer to the level in the intact group.
Conclusions. The experimental studies conducted allow us to recommend the use of domestic preparations of quertulin and propoxazepam in the clinic for the treatment and prevention of inflammatory and dysbiotic processes in the digestive tract in conditions of intoxication, in particular with potassium perchlorate.
